Molecular and cytogenetic alterations are involved in virtually every facet of acute myeloid leukemia (AML) cell transformation including dysregulation of major signal transduction pathways. In many instances, and in an ever growing numbers, these alterations define the leukemia subtype. Recent advances now provide flow cytometry techniques to sensitively measure multiple phosphorylated(p)-proteins coupled with lineage/differentiation related cellular antigens. These techniques offer rapid and specific measures of the signal pathway dysregulation that occurs in AML and may provide key adjunctive diagnostic data in AML as well as playing a role in therapeutic management of patients. The goal will be to show that the complex patterns observed in (Boolean gated) bivariate plots are derived from the cellular programmed expression of the parameters. We will use the cell cycle related expression of DNA, cyclins A2 and B1, and phosphorylation of histone H3 as a typical cell process. Using laser scanning cytometry data, we will integrate higher level information into a typical analysis. Space is limited.
